Summary:A study entitled "Random Amplified Polymorphic DNA (RAPD) analysis of banana (Musa spp.)" was conducted at the Department of Pomology and Floriculture, College of Agriculture, Vellayani, Thiruvananthapuram during the period from 2001 to 2003. Analysis of the genetic diversity. of some of the banana clones and intra-clones belonging to different genomic groups (AAA, AAB, ABB, AA, AB and BB) and ploidy groups (triploids and diploids), based on morphological traits and RAPD profiles is aimed at. Twenty eight banana clones were used for the study. Clones and intraclones of banana were collected from Banana Research Station (BRS), Kannara; Banana Nursery Farm, Palode, Thiruvananthapuram; Instructional Farm, College of Agriculture, Vellayani and Batlagundu, Tamil Nadu. DNA isolated from the twenty eight banana clones were subjected to RAPD analysis. Out of the forty one decamer primers screened for RAPD analysis, 34 could produce amplification. One hundred and twenty three bands (average: 3.00 bands per primer) were generated by thirty four primers, of which 94.31 per cent (116 bands) were polymorphic. Seven bands (5.69 %) were monomorphic. Twenty five primers showed high level of polymorphism. Finally, six most promising primers viz., OPA-Ol, OPA-03, OPA-13, OPB-04, OPB-IO and OPB-12 were used for RAPD analysis of the twenty eight banana clones. These primers yielded 46 scorable bands with an average of 7.66 bands per pnmer. From the dendrogram, it was found that the twenty eight banana clones clustered into fourteen groups at a distance of 0.200. At a distance of 0.250, 8 out of 12 Nendran (AAB group) intraclones viz., Chengazhikodan, Myndoli, Kaliethan, Vellayani Nendran, Zanzibar, Mysore Ethan, Manjeri Nendran and Changanasseri Nendran formed single cluster. Palayankodan (AAB group) intraclones like, PKNNR, Pisang Ceylon, Motta Poovan, Chandra Bale and Palode Palayankodan formed another single cluster. At a distance of 0.120, Red banana and Vellakappa (AAA group) as well as Pisang Ceylon and PKNNR (AAB group) formed single clusters. At a distance of 0.220, Kunnan and Njalipoovan (AB group) formed single cluster. Kadali and Pisang Lilin (AA group) as well as Quintal banana and Padalamurian (AAB group) formed single clusters at a distance of 0.240. Attu Nendran, Monthan, Robusta, Koonoor Ethan, Ilavazha and Vellapalayankodan formed individual clusters and had maximum genetic divergence. Among the diploid clones, Ilavazha (BB group) had maximum genetic divergence. Among the triploid clones, Attu Nendran, Robusta, Koonoor Ethan and Vellapalayankodan showed the maximum genetic divergence. Among the AAA group, Robusta recorded the maximum genetic divergence. Among the Nendran (AAB group) intraclones, Attu Nendran and Koonoor Ethan got maximum genetic divergence. Among the Palayankodan (AAB group) intraclones, Vellapalayankodan recorded the highest genetic divergence. In D2 analysis (based on morphological characters), Tocher's method was applied to develop six clusters. The Palayankodan (AAB group) intraclones like Palcde Palayankodan, PKNNR, Pisang Ceylon, Mottapoovan and Chandra Bale, AA group clones like Kadali and Pisang Lilin and AB group clones like Njalipoovan and Kunnan came under the first cluster. There were fourteen clones in the second cluster viz., Red banana, Vellakappa and Robusta (AAA group), Nendran (AAB group) intraclones like Padalamurian, Myndoli, Attu Nendran, Chengazhikodan, Kaliethan, Mysore Ethan, Manjeri Nendran and Changanasseri Nendran, ABB group clones like Monthan and Peyan and the BB group clone, Ilavazha. Vellayani Nendran and Zanzibar .(AAB group) came under the third cluster. Three clones (belonging to AAB group) remained independent, viz., Quintal banana (fourth cluster), Vellapalayankodan (fifth cluster) and Koonoor Ethan (sixth cluster). An analysis of the 28 banana clones with respect to 28 vegetative characters revealed that, M. acuminata triploids were more delicate. M. balbisiana triploids were more robust and hardy than Musa acuminata diploids, M. balbisiana, triploids and diploids were generally hardy in nature. With respect to yield characters, M. acuminata triploids were better than M. acuminata diploids. In M. balbisiana, both triploids and diploids were better yielder, compared to M acuminata. Among the M. acuminata, triploids yielded better than diploids. M. acuminata triploids had better quality than M. acuminata diploids. Among the M. balbisiana varieties, both diploids and triploids had good quality.
